Geobacillus CRISPR-Cas9 DescriptionA universal CRISPR_Cas9 gene editing system for broad application in thermophilic Parageobacillus and other anaerobic Gram +ve thermophiles, comprising 3 vectors. The widely used CRISPR/Cas9 nuclease of Streptococcus pyogenes is inactive at temperatures above 42 °C, making its use in thermophiles problematical. The three vectors included in the kit are:- pMTL575555 (stCas91), pMTL675555 (stCas93) and pMTL-RiboCas93. All vectors carry the ColEI and pUB110 origins of replication, a thermostable kanamycin resistance gene and a locus encoding the specific sgRNA while pMTL-RiboCas93 contains a Pgapdh RbxE riboswitch upstream of Cas9. All vectors are supplied at a concentration of 50ng/µl DNA are ready to be transformed into E. coli.
