Description

A selection of E. coli/Clostridium transposon shuttle vectors carrying a mariner element suitable for random mutant generation in various clostridial species and other Gram-positive bacteria. Those plasmids (pMTL-SC1, pMTL-YZ14, pMTL-CW20) using the PtcdB promoter require the presence of a chromosomal copy of tcdR, either natively present or inserted at the pyrE locus using ACE.

pMTL-SC1 – Exemplified in Clostridioides difficile, uses the pBP1 replicon and PtcdB promoter to express the transposase.

pMTL-YZ14 – Exemplified in Clostridium acetobutylicum, uses the pCB102 replicon and PtcdB promoter to express the transposase.

pMTL-CW20 – Exemplified in Clostridium autoethanogenum, uses the pCB102 replicon and PtcdB promoter to express the transposase.

pMTL-CW21– Exemplified in Clostridium sporogenes, uses the pCB102 replicon and the inducible Tet promoter in place of PtcdB , negating with the need for tcdR in the chromosome.

pMTL-CW22 - Exemplified in Clostridium sporogenes, uses pCD19 replicon and the inducible Tet promoter in place of the PtcdB promoter, negating the need for tcdR in the chromosome.

All vectors are supplied at a concentration of 50ng/µl DNA are ready to be transformed into E. coli.
