Vectors
Vectors needed for the genome engineering of Clostridium autoethanogenum based on its endogenous Type I-B CRISPR/Cas system.
Guidelines for vector design and transformation in Clostridium autoethanogenum will be provided. The four vectors supplied are as follows:
- pMTL-MPD25: empty cloning vector, for construction of KO and KI vectors containing the C. autoethanogenum leader sequence and a MCS for insertion of spacer and homology arms
- pMTL-MPD28: positive control for pyrE gene KO.
All plasmid maps will be provided in GenBank format.
All vectors are supplied at a concentration of 50 ng/µl DNA are ready to be transformed into E. coli.
