Vectors
Vectors needed for the genome engineering of Acetobacterium woodii based on its endogenous Type I-B CRISPR/Cas system.
Guidelines for vector design and transformation in Acetobacterium woodii will be provided. The four vectors supplied are as follows:
- pMTL-MPD15: empty cloning vector, for construction of KO and KI vectors. The empty cloning vector contains the A. woodii leader sequence and a MCS for insertion of spacer and homology arms.
- pMTL-MPD21: positive control for pheA KO.
- pMTL-MPD23: cloning vector for cargo delivery at the pheA locus; cargo to be inserted into the MCS.
- pMTL-MPD24: positive control for cargo delivery at the pheA locus; cargo is 614 bp and contains the FAST reporter gene under the control of the Pthl promoter.
All plasmid maps will be provided in GenBank format.
All vectors are supplied at a concentration of 50 ng/µl DNA are ready to be transformed into E. coli.
